Carbon-carbon bonding enzymes can be attractive alternatives to standard chemical methods by allowing chiral control, taking advantage of mild reaction conditions and minimizing the use of protecting groups in reactions. Fructose-6-phosphate aldolase (FSA) from Escherichia coli catalyses reversibly the cleavage of D-fructose-6-phosphate into dihydroxyacetone (DHA) and D-glyceraldehyde-3-phosphate (GAP). In addition, the enzyme creates new building blocks with 3S, 4R configuration by use of DHA as donor and various aldehydes as acceptor. The goal of this work was to investigate FSA and compare it with the transaldolase from Bacillus subtilis (TALBsu) which is similar both in sequence and structure to FSA (30% identical in amino acid residues). This should help to elucidate relationships between structure and function of both enzymes, and possible applications for FSA and TALBsu for the production of valuable sugars or sugar derivatives.

The oncogene v-myb of the retroviruses AMV (avian myeloblastosis virus) and E26 (avian leukaemia virus) encodes a transcription factor (v-Myb) which is a truncated homolog of its cellular progenitor c-Myb. c-Myb plays an essential role in the development of haematopoietic cells and is known to be a regulator for many target genes. v-Myb AMV is responsible for the transformation of myelomonocytic cells and for arresting them in an immature stage, presumably because of a deregulation of the expression of specific target genes. In addition to the truncation of the coding region, a number of amino acid substitutions are responsible for the high oncogenicity of v-Myb AMV. Due to the amino acid substitutions, v-Myb AMV and v-Myb E26 differ in their target gene spectrum. The chicken mim-1 gene is activated by v-Myb E26 and c-Myb but not by v-Myb AMV. The gene consists of two cis-regulatory regions, a Myb responsive promoter and a cell-specific Myb-inducible enhancer. Recently, it has been proven that two amino acid substitutions in a hydrophobic patch in the transactivation domain of v-Myb AMV are sufficient to disrupt its ability to stimulate the enhancer. This work focuses on the consequences of these amino acid substitutions by investigating protein-protein interactions of the hydrophobic region of v-Myb AMV in comparison to v Myb E26. Previous experiments identified GRP78 as an interaction partner of v Myb. In this study, a cytosolic variant of GRP78, GRP78va, was confirmed to interact with both v-Myb proteins. It was shown that its interaction site is limited to a very small region of v-Myb preceding the hydrophobic patch. Additionally, it was shown that GRP78va associated with all other members of the Myb-family and also with C/EBPß and HIPK2, suggesting a non-sequence-specific binding of GRP78va. Furthermore, reporter gene experiments demonstrated a repressing effect of GRP78va on the transactivation potential of v-Myb E26. In addition, GST pull down assays and co-immunoprecipitation experiments were used to precipitate endogenous proteins that could represent potential interaction partners of v-Myb. SDS-PAGE analysis revealed candidate bands, but mass spectrometry analysis failed to identify any proteins relevant for interaction with v Myb. Two other proteins were tested for their interaction with the hydrophobic patch of v-Myb. Co-immunoprecipitation experiments confirmed that C/EBPß interacts with the hydrophobic region of v-Myb and that the amino acid substitutions seem to affect the interaction in a negative way. Furthermore, PRMT4 was identified as an interaction partner of v-Myb. Mapping experiments showed the interaction to be mediated by the hydrophobic region. The point mutations in v-Myb AMV appear to positively influence the affinity for PRMT4. The fact that a SUMO binding motif is located in the same region might suggest a potential involvement of SUMO in the interaction of PRMT4 and v-Myb.

The oncogene v-myb of the retroviruses AMV (avian myeloblastosis virus) and E26 (avian leukaemia virus) encodes a transcription factor (v-Myb) which is a truncated homolog of its cellular progenitor c-Myb. c-Myb plays an essential role in the development of haematopoietic cells and is known to be a regulator for many target genes. v-Myb AMV is responsible for the transformation of myelomonocytic cells and for arresting them in an immature stage, presumably because of a deregulation of the expression of specific target genes. In addition to the truncation of the coding region, a number of amino acid substitutions are responsible for the high oncogenicity of v-Myb AMV. Due to the amino acid substitutions, v-Myb AMV and v-Myb E26 differ in their target gene spectrum. The chicken mim-1 gene is activated by v-Myb E26 and c-Myb but not by v-Myb AMV. The gene consists of two cis-regulatory regions, a Myb responsive promoter and a cell-specific Myb-inducible enhancer. Recently, it has been proven that two amino acid substitutions in a hydrophobic patch in the transactivation domain of v-Myb AMV are sufficient to disrupt its ability to stimulate the enhancer. This work focuses on the consequences of these amino acid substitutions by investigating protein-protein interactions of the hydrophobic region of v-Myb AMV in comparison to v Myb E26. Previous experiments identified GRP78 as an interaction partner of v Myb. In this study, a cytosolic variant of GRP78, GRP78va, was confirmed to interact with both v-Myb proteins. It was shown that its interaction site is limited to a very small region of v-Myb preceding the hydrophobic patch. Additionally, it was shown that GRP78va associated with all other members of the Myb-family and also with C/EBPß and HIPK2, suggesting a non-sequence-specific binding of GRP78va. Furthermore, reporter gene experiments demonstrated a repressing effect of GRP78va on the transactivation potential of v-Myb E26. In addition, GST pull down assays and co-immunoprecipitation experiments were used to precipitate endogenous proteins that could represent potential interaction partners of v-Myb. SDS-PAGE analysis revealed candidate bands, but mass spectrometry analysis failed to identify any proteins relevant for interaction with v Myb. Two other proteins were tested for their interaction with the hydrophobic patch of v-Myb. Co-immunoprecipitation experiments confirmed that C/EBPß interacts with the hydrophobic region of v-Myb and that the amino acid substitutions seem to affect the interaction in a negative way. Furthermore, PRMT4 was identified as an interaction partner of v-Myb. Mapping experiments showed the interaction to be mediated by the hydrophobic region. The point mutations in v-Myb AMV appear to positively influence the affinity for PRMT4. The fact that a SUMO binding motif is located in the same region might suggest a potential involvement of SUMO in the interaction of PRMT4 and v-Myb.

Accelerationism is the name of a contemporary political heresy: the insistence that the only radical political response to capitalism is not to protest, disrupt, critique, or détourne it, but to accelerate and exacerbate its uprooting, alienating, decoding, abstractive tendencies. #Accelerate presents a genealogy of accelerationism, tracking the impulse through 90s UK darkside cyberculture and the theory-fictions of Nick Land, Sadie Plant, Iain Grant, and CCRU, across the cultural underground of the 80s (rave, acid house, SF cinema) and back to its sources in delirious post-68 ferment, in texts whose searing nihilistic jouissance would later be disavowed by their authors and the marxist and academic establishment alike. On either side of this central sequence, the book includes texts by Marx that call attention to his own “Prometheanism;” and key works from recent years document the recent extraordinary emergence of new accelerationisms steeled against the onslaughts of neoliberal capitalist realism, and retooled for the twenty-first century. Er bietet vier Parts die sich gleichzeitig spielen lassen und insgesamt 16 Stimmen. Die Bedienung ist schnell verinnerlicht und das robuste Gehäuse vermittelt sofort Vertrauen. Auch klanglich zeigt sich die hohe Qualität: Die 24-Bit-DACs liefern einen sauberen detailreichen Output. Der Manatee steht offen zu seinem digitalen Wesen - und genau das macht seinen Sound aus. Er klingt eigenständig charmant und besitzt eine klare moderne Präsenz. Für eigene Klänge stehen 300 Speicherplätze bereit dazu 32 Multisets die komplette Vier-Part-Setups inklusive Panorama Pegel und Reverb speichern. Der Reverb selbst wirkt musikalisch warm und flexibel da mehrere Algorithmen zur Auswahl stehen. Mit vollständigem MIDI-Support lässt sich der Synth perfekt in DAWs oder Sequencer einbinden. Polyphoner Aftertouch (MPE) eröffnet zudem ein sehr ausdrucksstarkes Spielgefühl. Die digitale Klangerzeugung im Detail Die digitale Klangerzeugung für eine einzelne Stimme macht den Anfang mit dem Oszillator-Block der im Manatee Spectral Generator genannt wird. Dieser erzeugt einen wohlklingenden Synthese-Mix aus Virtual Analog Waveform und 2-Operator FM. Mit einer überschaubaren Anzahl von Parametern wird ein obertonreiches Spektrum geschaffen das vielseitig manipulierbar und vor allem modulierbar ist. Ergänzt wird der Spectral Generator durch den Sub-Oscillator der aufgrund verschiedener Wellenformen inklusive Super Saw und Super Puls zu weitaus mehr im Stande ist als einfach nur eine Oktave tiefer zu tönen. Wer möchte kann sogar eigene kurze Wellenformen importieren um die Klangerzeugung noch vielseitiger zu gestalten. Nicht fehlen darf ein Rauschgenerator; der im Manatee ist mit einem Lowpass/Highpass Filter im Klangsprektrum anpassbar und erlaubt eine Reduzierung der Samplerate bis dass nur noch Knistern Britzeln und Glitches zu hören sind. Das ist perfekt für atonale Sounds Perkussives und effektartige Klänge. Den Abschluss macht Ringmodulator der sich aus den beiden Oszillatoren speist und in gewohnter Manier unharmonische und metallische Klänge liefert.Der Algorithmus des resonanten Multimode Filters wurde auf Basis neuester Erkenntnisse analoger Modelling-Technologie neu geschrieben. Klanglich überzeugt das Filter mit einem sehr weichen und natürlichen Charakter und kann in Extremeinstellungen aber auch entsprechend färben. Funktionell bietet das Filter die Charakteristiken Lowpass Bandpass und Highpass in jeweils 12 und 24 dB an; zudem reagiert die Schaltung sehr gut auf schnelle Hüllkurven und LFO-Modulationen.Der letzte Bestandteil eines Sounds ist die Overdrive-Sektion. Hier geht es dem aufwendig generierten Sound an den Kragen. Die analog klingenden Verzerrer-Modelle reichen von leichter Sättigung über Röhren-Overdrive bis hin zu aggressiven Varianten wie Clipping und Bitcrush. Es profitieren wohl sanfte Pad Sounds aber auch harte Acid-Bässe von dem Möglichkeiten der Overdrive-Sektion.Letztendlich landen alle vier Parts in der Mixer-Sektion; hier warten ein Delay-Effekt mit Mono- Pingpong- oder Stereo-Mode einschließlich BPM-Sync ein Equalizer mit Cut- Boost- oder Shelf-Modi sowie der sehr gut klingende Insert Reverb nur darauf den Sounds den letzten Schliff zu geben.
